Your title page inside the book will have the inside 1/4" in the binding (approximately). I think that is negligible (possibly unnoticeable by the eye), but if you really want it centered, center it with from 1/4" in from the left and 1/8" in from the right, that's the approximate trim amount. I think if it's off by 1/8" you wouldn't notice. I centered my title page on the page itself and I can't notice what's lost in reference to the binding.

I should have asked how big the book will be, how many pages and what dimension, but I don't really think it matters. I always keep in mind that 1/4" on the gutter side of my pages through the book and it has always worked perfectly. On thicker books, farther into the book where there is more tension on the paper, 1/4" is barely enough, but nothing has ever been cut off.
I would hate to give you bad info---but the preview is really true to what you'll get. If it looks centered on the preview, you'll be okay. Hope it works!
